Clustering of Cx43 cell-to-cell channels into gap junction plaques: regulation by cAMP and microfilaments.

چکیده
Cell-to-cell channels are often seen clustered at cell-cell contacts into the so-called gap junction plaques. The mechanism of this clustering is unknown. We show that the clustering of cell-to-cell channels composed of connexin43 is induced by elevation of cyclic AMP. The cAMP-induced clustering is enhanced by inhibition of glycosylation and abolished by disruption of microfilaments. Channel clustering thus seems to be regulated by cAMP and glycosylation and to involve microfilaments.

The origin of annular junctions: a mechanism of gap junction internalization.
Gap junctional intercellular communication is established when connexin proteins oligomerize into connexon hemichannels, which then pair at the cell surface with connexons from neighboring cells to form functional gap junction channels. Gap junction channels routinely cluster into gap junction plaques, which can exhibit dynamic characteristics while under the frequent processes of formation and...
متن کاملGap Junction Channels Exhibit Connexin-specific Permeability to Cyclic Nucleotides
Gap junction channels exhibit connexin dependent biophysical properties, including selective intercellular passage of larger solutes, such as second messengers and siRNA. Here, we report the determination of cyclic nucleotide (cAMP) permeability through gap junction channels composed of Cx43, Cx40, or Cx26 using simultaneous measurements of junctional conductance and intercellular transfer of c...
متن کاملAtrial gap junction remodeling: looking for lost gaps and orphaned connexins in three dimensions.
Gap junction channels are composed of members of a multi-gene family of proteins called connexins. These proteins are named by the abbreviation Cx followed by the molecular weight of the specific protein. Mammalian cardiac myocytes express Cx43, Cx45, and Cx40, the former being the major cardiac gap junction protein, which is expressed in atrial and ventricular myocytes as well as in selected r...
متن کاملGap junction turnover, intracellular trafficking, and phosphorylation of connexin43 in brefeldin A-treated rat mammary tumor cells
Intercellular gap junction channels are thought to form when oligomers of connexins from one cell (connexons) register and pair with connexons from a neighboring cell en route to forming tightly packed arrays (plaques). In the current study we used the rat mammary BICR-M1Rk tumor cell line to examine the trafficking, maturation, and kinetics of connexin43 (Cx43). Cx43 was conclusively shown to ...
متن کاملAnchored PKA as a gatekeeper for gap junctions
Anchored protein kinase A (PKA) bound to A Kinase Anchoring Protein (AKAP) mediates effects of localized increases in cAMP in defined subcellular microdomains and retains the specificity in cAMP-PKA signaling to distinct extracellular stimuli. Gap junctions are pores between adjacent cells constituted by connexin proteins that provide means of communication and transfer of small molecules. Whil...
